Use your putty knife and apply the asphalt flashing cement to the back of the piece you just cut or your patch.
If you have a gutter patch you can use it.
Make sure your primer is ammonia free as a primer with ammonia could react with the aluminum causing gas bubbles to occur between the layers.
If not cut a patch from the metal flashing that is just a bit bigger than your hole with your metal snips.

If you are not comfortable working on heights the best solution is to remove the gutter section by section and paint it on the safety of your grounds.

Leave the gutters where they are only if you feel 100 comfortable and have performed a diy job before.
If the old paint is really caked onto the gutters you can use paint stripping.
